What is Imbruvica 140mg (ibrutinib)?

Imbruvica 140mg (ibrutinib) is used to treat people with mantle cell lymphoma (MCL; a fast-growing cancer that begins in the cells of the immune system) who have already been treated with at least one other chemotherapy medication. Imbruvica 140mg (ibrutinib) is also used to treat people with chronic lymphocytic leukemia (CLL; a type of cancer that begins in the white blood cells) and small lymphocytic lymphoma (SLL; a type of cancer that begins mostly in the lymph nodes). Ibrutinib is used to treat people with Waldenstrom’s macroglobulinemia (WM; a slow-growing cancer that begins in certain white blood cells in your bone marrow).

Administration

Oral Administration

Administer once daily at approximately the same time each day

Swallow capsule whole, do not chew, break, open, or crush capsules

When given in combination with rituximab or obinutuzumab, consider administering ibrutinib prior to rituximab or obinutuzumab when given on the same day

Missed dose

  • Take missed dose as soon as possible on the same day and return to normal schedule the following day
  • Do not take extra capsules to make up for the missed dose

Storage

Store at room temperature between 68-77°F (20-25°C)

Keep ibrutinib in the original container with the lid tightly closed.

Precautions:

  • Do not drink grapefruit juice, eat grapefruit or eat Seville oranges (often used in marmalades) while you are taking Imbruvica. These products may increase the amount of Imbruvica in your blood.
  • Before starting Imbruvica treatment, make sure you tell your doctor about any other medications you are taking (including prescription, over-the-counter, vitamins, herbal remedies, etc.).
  • While taking Imbruvica, do not receive any kind of immunization or vaccination without your doctor’s approval.
  • Limit your time in the sun. Imbruvica can make your skin sensitive to sunlight. A severe sunburn, rash or worsening acne can occur with too much exposure. Remember to use sunscreen and wear a hat and clothes that cover as much of your skin as possible while taking Imbruvica.
  • Inform your health care professional if you are pregnant or may be pregnant prior to starting this treatment. Pregnancy category D (may be hazardous to the fetus. Women who are pregnant or become pregnant must be advised of the potential hazard to the fetus.)
  • For both men and women: Barrier methods of contraception, such as condoms, are recommended during therapy and for at least 2 weeks after treatment is complete. Discuss with your doctor when you may safely become pregnant or conceive a child after therapy.
  • Do not breast feed while taking Imbruvica.

Side effects Imbruvica 140mg (ibrutinib)

Imbruvica (ibrutinib) is an inhibitor of Bruton’s tyrosine kinase (BTK) used to treat patients with mantle cell lymphoma (MCL) who have received at least one prior therapy. Common side effects of Imbruvica include:

  • low platelet count,
  • diarrhea,
  • neutropenia,
  • anemia,
  • fatigue,
  • musculoskeletal pain,
  • muscle spasms,
  • joint pain,
  • swelling of the extremities,
  • fever,
  • upper respiratory tract infection,
  • cough,
  • shortness of breath,
  • nausea,
  • bruising,
  • indigestion,
  • constipation,
  • rash,
  • abdominal pain,
  • vomiting,
  • decreased appetite,
  • swelling of the mouth and lips,
  • urinary tract infection,
  • pneumonia,
  • skin infections,
  • sinus infection,
  • weakness,
  • nosebleed,
  • decreased appetite,
  • dehydration,
  • dizziness, and
  • headache.

The recommended dose of Imbruvica for MCL is 560 mg (four 140 mg capsules) orally once daily. Imbruvica may interact with ketoconazole, itraconazole, voriconazole, posaconazole, clarithromycin, telithromycin, grapefruit and oranges, carbamazepine, rifampinphenytoin and St. John’s Wort. Tell your doctor all medications and supplements you use. Imbruvica is not recommended for use during pregnancy. It can harm a fetus. It is unknown if this drug passes into breast milk.
